The Argentina international has been ruled out for four to five weeks by a hamstring strain.
Spurs boss Ange Postecoglu confirmed the news at his press conference this morning.
Romero left the win over Everton on Saturday at half-time.
Left-back Destiny Udogie returns from suspension.
Emerson Royal, who played at left-back in Udogie’s absence, and Eric Dier appear candidates to fill in for Romero.
James Maddison, Manor Solomon and Ivan Perisic are among Tottenham's absentees but they have recalled striker Dane Scarlett from his loan at Ipswich.
Albion head coach Roberto De Zerbi will speak to the media this afternoon.
