Motorists face a lengthy detour as roadworks mean part of the westbound A27 will be closed overnight.
The resurfacing works will take place between the junction with the B2123 The Drove and the A270 Lewes Road from 8pm and 6am tonight.
Drivers heading west will be forced to head south along the Drove before cutting through Warren Road and Bear Road.
The diversion continues onto Lewes Road and heading north onto Coldean Lane to the Hollingbury interchange.
The Highways Agency said the work had been postponed from May 9 due to bad weather but had chosen overnight to minimise disruption.
Access to and exit from Falmer station will be maintained.
A Highways Agency spokesperson said: "The work being carried out tonight on the A27 at Falmer Hill involves essential resurfacing of the carriageway.
"We would usually provide as much notice as possible . However on this occasion the work needs to be done as soon as possible, and is not a planned scheme.
"The work had to be postponed from last week , when it was originally announced, due to adverse weather conditions."
The project is one of several happening in May along the A27, including work to the Southwick tunnel and Southerham railway bridge.
