Business organisations fear the Government will continue its inaction on the increasingly congested A27.
They are concerned serious traffic problems are damaging business in the area.
The road's poor design has also been blamed for numerous accidents.
Most recently, three students died in a crash on the notorious road.
The Government rejected proposals to improve the A27 in 2003, leaving businesses along the route feeling left out.
Chief executive of Sussex Enterprise Mark Froud wrote to Dr Stephen Ladyman, Minister of State for Transport.
He said: "The Sussex road network is in desperate need of improvements. Unless they are made, not only will businesses be deterred from investing in the area, but some businesses may be forced to relocate from Sussex."
The Regional Transport Board of SEERA, the South East England Regional Assembly, has given a low ranking to schemes for bypasses along the A27.
This means they have not been identified for delivery until after 2011 at the earliest.
