Campaigners against a directly-elected mayor were jubilant after hearing the latest result from another city.
Sunderland in the North-East voted decisively against having a mayor in a referendum on a low turnout.
Councillor Brian Oxley, Tory opposition leader on Brighton and Hove Council, said: "This result, following on from others, is showing a clear trend against elected mayors.
"People are showing they feel that it is not constitutional upheaval that will make their lives better but a genuine focus on problem-solving in their localities by the local representatives they currently have."
The Brighton and Hove referendum is being carried out now on a postal ballot with the result being announced on Friday.
