Liam Barham and John Crumplin are surprise inclusions in Lewes's squad for their showdown with AFC Sudbury at the Dripping Pan.
St Leonards skipper Barham is expected to take over from broken leg victim Justin Harris in midfield while ex-Albion defender Crumplin is likely to be a substitute.
Both players have agreed to help a club ravaged by injury, suspension and the loss of cup-tied players.
Barham's appearance will be a one-off as part of an arrangement Lewes have with St Leonards. Crumplin, who started the season at St Leonards, has been playing for Three Bridges in the County League.
Rooks are without cup-tied Julian Curnow, Germain Amanuel, Mackay and Mike Ebanks, although the latter's loan spell is about to end.
In addition to Harris, Paul Stokes and Ross Venables are injured while Hack collected an ankle injury playing for St Leonards last weekend.
Joe Clark, who would have taken over from Harris, is suspended and Artur Correia has left the club.
Peter Brackley is another long term injury casualty with a broken wrist but Quinn may be forced to name him as a substitute as he did in the last round against Tow Law Town.
Two teenage reserve players will make up the numbers in a squad which also includes recent signing Clinton More.
Quinn said: "We will have to do it the hard way again.
"To the players' credit they have always pulled out big performances and they will have to do it again tomorrow if we are to have any chance.
Lewes have erected a temporary stand which will stay up for the rest of the season. Under-14s can get in for free and more than 300 tickets have also been sent out to local schoolchildren.
AFC Sudbury are currently top of the Jewson Eastern Counties League.
