Sean Baldock failed in his bid to claim the third and final 400m place in the Great Britian Olympic squad, which will be announced today.
The Hastings-based runner - who competed in the Sydney Olympics four years ago - could only manage a time of 46.35sec in Madrid on Saturday night, when he needed to break 45.55sec.
That was only good enough for seventh place in the IAAF Super Grand Prix meeting and Baldock will now have to be satisfied with a place in the 4x400m relay team for Athens.
American-born Malachi Davis is expected to take the third individual spot despite finishing behind Baldock in the Olympic trials eight days ago.
Baldock's coach Mark Gregory said: "Sean is obviously very disappointed but he is in the relay so we must try to focus on the positives.
"It was a good race and everything was set up well but Sean just didn't feel right. There are no excuses it was just one of those things."
Mick Jones will also be missing from the squad after failing to make the B standard of 74.35m in the hammer at Eton yesterday.
The British No. 1 from Crawley won the hammer to help his club Belgrave reach the final of the British League Cup but fell short of the qualifying mark.
