Darren Mateer, 30, from Whitehawk, is accused of murder and affray.
His brother Jonathan, 26, also from Whitehawk, is charged with affray.
A19-year-old woman arrested after the death of Jay Kensett, 16, has since been released on bail.
Jay died at the Royal Sussex County Hospital, Brighton, after suffering a stab wound in Whitehawk Way shortly before midnight on Friday.
Police later recovered a knife.
More flowers and tributes were left today at the scene of Jay's death, one from his fiancee, Cassie Watts.

Jay's parents John and Kim, both 40, from Pulborough Close, Whitehawk, watched as attempts were made to revive him.
The Marina High School pupil was a talented boxer and footballer.
He had been out celebrating after scoring four goals for his school.
Whitehawk FC's first team dedicated its Saturday match against Shoreham to Jay, and more than 100 fans held a minute's silence before the kick-off.
