Residents are opposing the transfer of the Royal Alexandra Children's Hospital to the site of the Royal Sussex County Hospital.
People in Turton Close, off Whitehawk Hill Road, Brighton, believe the redevelopment of the children's hospital will impinge on their view, create traffic, noise and safety problems, and devalue their properties.
They also fear the redevelopment of the kidney unit at the Royal Sussex will have similar effects.
Resident Joanna Everall said: "The building work will cause disruption just like we have had for the last few years while they have been redeveloping other parts of the hospital.
"It will also be difficult to get into our little close and there will be problems with traffic getting in and out. It would be better to relocate the hospital elsewhere."
The new children's hospital has not yet received planning permission from Brighton and Hove City Council and the residents plan to object to it.
John Hopkins, who lives in a sixth-floor flat in Turton Close, said: "We used to be able to see the Palace Pier from our flat. But we can only see part of it now because of new hospital buildings. After the redevelopment our view will get worse again."
A spokesman for Brighton Health Care NHS Trust, which runs the Royal Sussex and Royal Alex, said: "We completely understand the concerns they have got and we want to continue to do everything we can to minimise the disruption to our neighbours.
"In order to develop excellent hospital facilities for the new city of Brighton and Hove there will inevitably be some disruption.
"Ultimately we believe that a fantastic 21st Century hospital will benefit everyone and perhaps lead to an increase in the value of their homes."
