Two County League sides collected seven-goal hauls to march into the next round in impressive style.
Sidlesham marked their debut in the competition by beating Redhill 7-0 while Broadbridge Heath, of division two, sunk Whitchurch United by the same margin.
Jamie Liddell (2), Russell Harwood, Ramin Jarvand, Stuart McCall, Paul Gallagher and Warren Sweetman scored Heath's goals against the Wessex League side who had a player sent off midway through the second half.
Heath manager Sam Chapman said: "We were 3-0 up after 15 minutes and they were always playing catch-up after that.
"We were expecting a tough game, both physically and in quality, but we had a good day and killed them off."
Sidlesham inflicted an embarrassing defeat on Sussex rivals Redhill. Richie Davies grabbed four goals, his prolific team-mate Scott Tipper bagged a brace and there was one from substitute Craig Williams which flew in from 25 yards.
Manager Richard Towers said: "It was a pleasing all-round team performance and Davies and Tipper have got about ten or 11 apiece this season so we are all happy.
"It is nice to win especially as it was our first game in the FA Vase in the club's history."
Whitehawk's in-form marksman Tommy Pattenden was on the scoresheet and Terry Cooper also found the net as they beat Reading Town, of the Combined Counties League, 2-0 in comfortable fashion with a good first-half display.
Hawks boss Ian Chapman said: "Reading did not have a real chance but we fell away a bit in the second half."
Hailsham won 1-0 at Lancing after Paul Spencer followed up a blocked shot by Scott French to force the ball home.
Winning manager Brian Dennis was pleased his side kept a cleansheet for the first time this season.
Withdean 2000, of the Combined Counties League, beat Ryman outfit Camberley Town 2-0 with Sam Francis netting a brace with goals in either half.
Withdean chief Alan Pook said: "We were always in control and should have won by more but missed a number of chances. Jay Pickering, Ollie Rowland and Dominic Shepherd should have scored.
"It is nice to go away from home and win cup games in style. We made sure of the result."
East Grinstead celebrated only their second Vase success in 15 years but not before a few worrying moments at home to a decent Herne Bay side.
Wasps won 4-3 but the game was held up for 35 minutes when Herne Bay's Sam Denly suffered a broken leg.
Wasps came from 2-1 down to lead 4-2.
A Ben Burns flick made it 2-2 and Phil Thompson got the fourth from 20 yards. Chris Arrow netted twice. Boss Bob Smith said: "We scored some great goals."
Arundel's finishing was also pretty good as they beat Hassocks 3-1 at Mill Road.
Centre forward Gary Norgate produced two great strikes and and had a third ruled out before Miles Scerri's penalty made it safe.
Anthony Hibbert sparked a Hassocks fightback in the closing stages and home goalkeeper James Latter had to make two superb double saves to keep his side in command.
New boss Richie Reynolds has plenty of work to do at Wick as his side were thrashed 11-1 at Lymington and New Milton's Fawcett's Field.
Wes Tate was on target at AFC Totton as Sidley lost 2-1 to the Jewson Wessex League outfit.
There were also defeats for Eastbourne Town, Ringmer, Southwick, Oakwood, Saltdean and Littlehampton.
