The police said the westbound carriageway was reopened, but it is understood the crash took place on the eastbound carriageway where motorists may still be affected by delays.
Sussex Police said: “We are in attendance at a road traffic collision on the A27 between Falmer and the Ashcombe Roundabout.
“The eastbound carriageway remains closed however the westbound carriageway has since re-opened.
“We are advising motorists to avoid the area and seek an alternative route.”
Images from the A27 this morning (Saturday, May 1) show long queues of vehicles on the eastbound carriageway already.
One passenger who contacted The Argus said a horse was being moved from one trailer to another.
Traffic was static on the dual carriageway, with reports of tailbacks for at least two miles.
Accident investigation work is underway and vehicles are being recovered.
